Ipamorelin

Ipamorelin is a selective growth hormone secretagogue that activates the ghrelin receptor (GHSR1A) in the pituitary gland. Unlike older GH secretagogues, Ipamorelin produces clean GH pulses without elevating cortisol, prolactin, or ACTH — making it the most selective compound in its class. It works through a completely different receptor pathway than CJC-1295, and the two are frequently used together for compounded GH stimulation.

How it works.

Ipamorelin is a pentapeptide that mimics ghrelin — the hunger hormone produced in the stomach — at its receptor in the pituitary gland (GHSR1A). When this receptor is activated, the pituitary releases a pulse of growth hormone. The key distinction: Ipamorelin activates only this receptor, unlike GHRP-2 and GHRP-6 which also activate receptors that raise cortisol and prolactin as side effects.

The GH pulse produced by Ipamorelin follows the body's natural rhythm and is dose-dependent. Timing is critical: administration before sleep aligns the peptide-induced pulse with the body's largest natural GH release, which occurs during slow-wave sleep. This produces a compounded effect without disrupting the feedback axis.

Downstream effects of sustained GH elevation include increased IGF-1 production, satellite cell activation in muscle, improved fat oxidation through lipolysis, enhanced collagen synthesis, and strengthened immune function via NK and T-cell activation.
